Provided is a fluorescent probe for detection in biological systems. Example compd. AJ2NBD, was prepd. from the reaction of 1,6-dimethyl-1,4,7,10-tetraazacyclododecane with 4-chloro-7-nitrobenzo[1,2,5]oxadiazole affording the desired compd. as a microscryst. solid in 80% yield. AJ2NBD was found to be compatible when used simultaneously with other probes such as Lysotracker Deep RED, Mitotracker Red, Dil, Bodipy, in that each probe was able to mark specific cell organelles such as mitochondria and lysosomes without interference. AJ2NBD was evaluated in serum-starved HT22 and HeLa cells and was found to brilliantly mark the perinuclear area indicating its usefulness in imaging vesicular trafficking.
